Tetracaine hcl of Method name:
Determination of Tetracaine hcl-Neutralization Titration Method
Application scope:
This method uses titration to determine the content of Tetracaine hcl.
This method is applicable to Tetracaine hcl.

Method principle:
After adding the test solution of glacial acetic acid and acetic anhydride, heat to reflux for 2 hours, let cool, add mercury acetate test solution and crystal violet indicator solution, titrate with perchloric acid titrant (0.1mol/L) until the solution is blue Color, record the amount of perchloric acid titrant used, calculate, and get it.
Reagents:
1. Water (return to room temperature)
2. Perchloric acid titrant (0.1mol/L)
3. Crystal Violet Indicator Solution
4. Acetic anhydride
5. Glacial acetic acid
6. Mercury acetate test solution
7. Benchmark potassium hydrogen phthalate
equipment:
Sample preparation:
1. Perchloric acid titration solution (0.1mol/L)
Preparation: Take 750 mL of anhydrous glacial acetic acid (calculated based on the water content, add 5.22 mL of acetic anhydride per 1 g of water), add 8.5 mL of perchloric acid (70%-72%), shake well, and slowly add acetic anhydride at room temperature 23mL, shake while adding, shake well after adding, let cool, add an appropriate amount of anhydrous glacial acetic acid to make 1000mL, shake well, and leave for 24 hours. If the tested product is easy to acetylate, the water content of this page must be determined by the water content method, and then adjusted with water and acetic anhydride to the water content of the solution to be 0.01%-0.2%.
Calibration: Take about 0.16g of standard potassium hydrogen phthalate dried at 105℃ to constant weight, accurately weigh it, add 20mL of anhydrous glacial acetic acid to dissolve, add 1 drop of crystal violet indicator solution, and slowly titrate with this solution to Blue, and the titration results are corrected with a blank test. Each 1mL perchloric acid titrant (0.1mol/L) is equivalent to 20.42mg of potassium hydrogen phthalate. According to the consumption of the liquid and the dosage of potassium hydrogen phthalate, calculate the concentration of the liquid.
Storage: Put it in a brown glass bottle and keep it tightly closed.
2. Crystal Violet Indicator Solution
Take 0.5 g of crystal violet, add 100 mL of glacial acetic acid to dissolve it, and get it.
Steps:
Accurately weigh 0.2g of the test product, add 25mL of glacial acetic acid and 5mL of acetic anhydride to dissolve, heat to reflux for 2 hours, let cool, add 5mL of mercury acetate test solution and 1 drop of crystal violet indicator solution, and use perchloric acid titration solution (0.1 mol/L) The titration solution is blue, and the titration result is corrected with a blank test, and the volume of perchloric acid titrant consumed (mL) is recorded. Each 1mL perchloric acid titrant (0.1mol/L) is equivalent to 30.08 mg of Tetracaine hcl (C15H24N2O2 ·HCl), that is.
